We present a model of excitable media with the feature that it has a vulnerable phase during which a premature current stimulus will result in the formation of a reentrant selfsustained wave of excitation. The model exploits anisotropic coupling of identical cells, and is therefore useful as a model for the myocardium. We give rigorous verification that there is a vulnerable phase, and demonstrate numerically that permanently rotating waves are formed. Finally, it is shown that the direction of fastest propagation in myocardium is not necessarily the direction of highest safety factor, contrary to commonly accepted opinion.  相似文献

A simple electrochemiluminescence-based assay for RNA N-glycosidase activity has been modified to permit its use with authentic extracts of Ricinus communis (castor beans) and Abrus precatorius (jequirity seeds)—the natural sources of ricin and abrin. Modifications include the addition of an RNase inactivator to the reaction mixture, elimination of a signal-enhancing monoclonal antibody, and optimization of the incubation temperature. Concurrent testing with two substrates provides a diagnostic tool enabling castor bean toxins to be differentiated from a larger selection of N-glycosidase toxins than was previously examined.  相似文献

Cardiac cells are electrically coupled through gap junction channels, which allow ionic current to spread intercellularly from one cell to the next. In addition, it is possible that cardiac cells are coupled through the electric potential in the junctional cleft space between neighboring cells. We develop and analyze a mathematical model of two cells coupled through a common junctional cleft potential. Consistent with more detailed models, we find that the coupling mechanism is highly parameter dependent. Analysis of our model reveals that there are two time scales involved, and the dynamics of the slow subsystem provide new mathematical insight into how the coupling mechanism works. We find that there are two distinct types of propagation failure and we are able to characterize parameter space into regions of propagation success and the two different types of propagation failure.  相似文献

Defibrillation of cardiac tissue can be viewed in the context of dynamical systems theory as the attempt to move a dynamical system from the basin of attraction of one attractor (fibrillation) to another (the uniform rest state) by applying a stimulus whose form is physically constrained. Here we give an introduction to the physical mechanism of cardiac defibrillation from this dynamical perspective and examine the role of resistive inhomogeneity on defibrillation efficacy. Using numerical simulations with rotating waves on a one-dimensional periodic ring, we study the role of the spatial scale of resistive inhomogeneity on defibrillation. For a rotating wave on a periodic ring there are three stable attractors, namely the uniform rest state, a wave traveling clockwise and a wave traveling counterclockwise. As a result, the application of a stimulus has the potential for three different outcomes, namely elimination of the wave, phase resetting of the wave, and reversal of the wave. The results presented here show that with resistive inhomogeneities of large spatial scale, all three of these transitions are possible with large amplitude shocks, so that the probability of defibrillation is bounded well below one, independent of stimulus amplitude. On the other hand, resistive inhomogeneities of small spatial scale produce a defibrillation threshold that is qualitatively consistent with that found experimentally, namely the probability of defibrillation success is an increasing function that approaches one for large enough stimulus amplitude. Extending these results to higher dimensions, we describe conditions for successful defibrillation of functional reentry with large scale spatial inhomogeneity, but find that elimination of anatomical reentry is quite difficult. With small spatial scale inhomogeneity, there are no similar restrictions.  相似文献

We present a mathematical model for the growth and length regulation of the hook component of the flagellar motor of Salmonella typhimurium. Under the assumption that the molecular constituents are translocated into the nascent filament by an ATP-ase and then move by molecular diffusion to the growing end, where they polymerize into the growing tube, we find that there is a detectable transition from secretion limited growth to diffusion limited growth. We propose that this transition can be detected by the secretant FliK, allowing FliK to interact with FlhB thereby changing the secretion target of the type III secretion machinery and terminating the growth of the hook.  相似文献

Few studies have examined the natural complex pigmentation patterns of white‐beaked dolphins. From 2002 to 2014, whale‐watching trips in Iceland provided a platform of opportunity to collect a large body of photographs of free‐ranging individuals from a single area of distribution for this species. Based on 823 images, 571 individuals showing one or more color components were identified, and assigned to the following four age classes: adults (n = 437), juveniles (n = 109), calves (n = 14), and neonates (n = 11). A total of 26 color components were observed and described: seven terms previously applied to white‐beaked dolphins, 12 previously applied to other dolphin species, and seven newly defined terms. Results showed that each age class could be positively identified by differences in specific color components, some of which were exclusive. Therefore, color patterns may prove useful in estimating maturity in free‐ranging white‐beaked dolphins. This tool could be further refined through assessment of a wide sample of freshly stranded specimens of known sex and age, which could reveal new age class‐specific components, as well as sexually dimorphic characteristics not seen here. Geographic variation should be investigated by comparing image data sets and stranded animals from different parts of the North Atlantic.  相似文献

Forward head and rounded shoulder posture (FHRSP) is theorized to contribute to alterations in scapular kinematics and muscle activity leading to the development of shoulder pain. However, reported differences in scapular kinematics and muscle activity in those with forward head and rounded shoulder posture are confounded by the presence of shoulder pain. Therefore, the purpose of this study was to compare scapular kinematics and muscle activity in individuals free from shoulder pain, with and without FHRSP. Eighty volunteers were classified as having FHRSP or ideal posture. Scapular kinematics were collected concurrently with muscle activity from the upper and lower trapezius as well as the serratus anterior muscles during a loaded flexion and overhead reaching task using an electromagnetic tracking system and surface electromyography. Separate mixed model analyses of variance were used to compare three-dimensional scapular kinematics and muscle activity during the ascending phases of both tasks. Individuals with FHRSP displayed significantly greater scapular internal rotation with less serratus anterior activity, during both tasks as well as greater scapular upward rotation, anterior tilting during the flexion task when compared with the ideal posture group. These results provide support for the clinical hypothesis that FHRSP impacts shoulder mechanics independent of shoulder pain.  相似文献

Important mechanical events during mitosis are facilitated by the generation of force by chromosomal kinetochore sites that attach to dynamic microtubule tips. Several theoretical models have been proposed for how these sites generate force, and molecular diffusion of kinetochore components has been proposed as a key component that facilitates kinetochore function. However, these models do not explicitly take into account the recently observed flexibility of kinetochore components and variations in microtubule shape under load. In this paper, we develop a mathematical model for kinetochore-microtubule connections that directly incorporates these two important components, namely, flexible kinetochore binder elements, and the effects of tension load on the shape of shortening microtubule tips. We compare our results with existing biased diffusion models and explore the role of protein flexibility inforce generation at the kinetochore-microtubule junctions. Our model results suggest that kinetochore component flexibility and microtubule shape variation under load significantly diminish the need for high diffusivity (or weak specific binding) of kinetochore components; optimal kinetochore binder stiffness regimes are predicted by our model. Based on our model results, we suggest that the underlying principles of biased diffusion paradigm need to be reinterpreted.  相似文献
